Our recent study published in Nature Communications has been featured by Asia Research News. The study reveals that O-linked glycans act as a “sugar brake” that suppresses the activity of the lysosomal ion channel TPC2. Disruption of this glycosylation-dependent regulation leads to TPC2 hyperactivation and alters lysosomal ion homeostasis and organelle morphology. By combining whole-lysosome patch-clamp electrophysiology, cell biology, and international collaboration, this work uncovers a previously unrecognized mechanism regulating TPC2 activity.

Special thanks to the first authors, Chien-Ju Alice Lin and Dr. Neng-Yu Lin, for their major contribution to this study, and to all co-authors, international collaborators, partner institutions, and funding agencies for their support.
